Production and Characterization of Putative Intertribal Somatic Hybrids Between Salpiglossis and Petunia

Abstract

Somatic hybridization was carried out between cytoplasmic albino cell suspension protoplasts of Salpiglossis sinuata and mesophyll protoplasts of Petunia hybrida cv. Monsanto and P. parodii. PEG/high pH fusion was effective in both combinations, not only for inducing a high fusion frequency but also for the recovery of hybrid calli. Somatic hybrid selection schemes were based on genetic complementation of homozygous recessive albino mutants as well as physiological complementation based on in vitro growth. Somatic hybrid calli were successfully obtained from both combinations. Shoot regeneration from hybrid calli was frequent, but root formation was limited. Morphological characteristics, chromosome numbers and isozyme banding patterns of selected regenerants indicated their somatic hybridity.
